3D-structural clustering of somatic mutations
An algorithm for identifying 3D clusters of somatic mutations on protein structures. By projecting mutations from large cancer cohorts onto solved and predicted protein structures, CLUMPS2 detects spatial hotspots of mutation enrichment that point to functionally relevant domains, complementing linear (1D) significance methods. It has been used to identify structurally clustered driver mutations in chronic lymphocytic leukemia in (Knisbacher et al., 2022) and in clonal hematopoiesis and myelodysplastic syndromes in (Beauchamp et al., 2021).
